Abstract Title Mircroarray Analysis of Mouse Limb Bud Ectoderm.

Text of abstract Mouse limb buds on E9 or 10 were excised from the flank, and lightly trypsinized to separate ectoderm from mesoderm. RNA was prepared from a pool of 25-40 ectoderms and hybridized to Affymetrix oligonucleotide chips. Samples of forelimb and hindlimb ectoderm were collected at two developmental time points when the limbs were ca. Stage 1.5 or 2.5-3.0. The forelimb samples were run on the Affymetrix U74A chip containing about 12,000 genes. The hindlimb samples were hybridized to the newest Affymetrix chip, MOE430 v2.0, containing the entire mouse genome. The results presented will focus on transcription factors and growth factors to present a universal view of these crucial molecular determinants of tissue differentiation. Also, focus on important signaling pathway, including Wnt, Hh, and Bmp, will demonstrate the presence, and intensity of pathway components, and how these change as development proceeds.
